The method involves acquiring a three-dimensional image for each of a set of signals emitted by a tissue e.g. melanin, where the image is acquired by performing multi-photon excitation of the tissue. A segmentation of the tissue is determined in two areas by processing of the three-dimensional image. The areas of the tissue are delimited, and a set of markers corresponding to each area is calculated. A propagation of the set of markers is calculated by utilizing a watershed algorithm. A graph is generated based on position of the set of markers. An independent claim is also included for a method for evaluating action of a stimulus and/or treatment.
