USE OF TAQ POLYMERASE MUTANT ENZYME FOR DNA AMPLIFICATION IN THE PRESENCE OF PCR INHIBITORY SUBSTANCE

PROBLEM TO BE SOLVED: To provide a DNA polymerase resistant to PCR inhibitory substances, a method of use of the polymerase in gene detection by standard and real-time PCR both in the presence and absence of blood and soil in the samples, as well as a method of using the polymerase in real-time assay in the presence of dye.SOLUTION: There are provided (i) a polypeptide sequence comprising a specific sequence, or (ii) an isolated polypeptide comprising a polypeptide sequence having at least 95% sequence identity to the polypeptide sequence having asparagine (N) at position 708, and resistant to a PCR inhibitory substance which is selected from the group consisting of dye, soil and blood or a combination thereof. There is also provided a method to amplify target nucleic acid by polymerase chain reaction (PCR) using a polymerase which is said isolated polypeptide.
