The present invention provides a recombinant herpes simplex virus and the preparation method thereof, wherein ICP4 gene promoter in herpes simplex virus genome including ICP4 gene was replaced by human telomerase reverse transcriptase promoter hTERTp or other tumor-specific promoters, so as to grow and increase in human tumor cells more selectively and killed cancer cells effectively. The present invention also provides an application of the recombinant herpes simplex virus for manufacturing a medicament for the treatment of tumor. The present invention also provides another recombinant herpes simplex virus, in which a fluorescent protein expression box was inserted to the said recombinant herpes simplex virus, so that the virus can emit light in tumor cells in order to achieve early diagnosis and diagnosis of tumor metastasis.
