The present invention is a gene sequence and a protein sequence of a pinewood nematode bursaphelenchus xylophilus expansin, a diagnostic composition and kit for pine wilt disease, and a pharmaceutical composition to prevent and treat pine wilt disease using the same. According to the present invention, a nevel expansin gene and protein sequence can be found from the pinewood nematode bursaphelenchus xylophilus causing the pine wilt disease. In addition, a medicine or a method to prevent and treat the pine wilt disease can be provided by studying an expression pattern and structure of the expensing protein. The present invention includes a pinewood nematode bursaphelenchus xylophilus expansin B gene having a nucleic acid sequence of SEQ ID NO: 25; and a diagnostic composition for pine wilt disease containing the pinewood nematode bursaphelenchus xylophilus expansin B gene having a nucleic acid sequence of SEQ ID NO: 25.
