The present invention relates to an A549 based cell line for preparing an adenovirus vector, and more specifically to an adenovirus preparation cell line of A549 based cell line containing adenovirus derived E1a gene and E1b gene. According to the present invention, there is an effect of providing the safe adenovirus preparation cell line having excellent efficiency of adenovirus preparation and low possibility of wild type virus infection.
