The present invention relates to a pharmaceutical composition for preventing or treating K-Ras-overexpressed cancer or suppressing the metastasis of K-Ras-overexpressed cancer, the composition containing: (a) a pharmaceutically effective amount of 5-bromo-3-(3-hydroxyprop-1-ynyl)-2H-pyran-2-one, or a salt, hydrate, or solvate thereof, as an active ingredient; and (b) a pharmaceutically acceptable carrier. The composition of the present invention can effectively perform prevention, treatment, or metastasis suppression of cancer cells which have resistance to the existing radiotherapy and chemotherapy and have excellent migrating and invading characteristics due to the overexpression of K-Ras.
