The present invention relates to a persimmon leaf-derived polysaccharide fraction with immunopotentiating activity and antitumor activity and a producing method thereof, and, more specifically, to a persimmon leaf-derived polysaccharide fraction with immunopotentiating activity and antitumor activity, a producing method thereof, and use thereof. The persimmon leaf-derived polysaccharide fraction comprises 60-80 wt% of neutral sugar, 18-39 wt% of uronic acid, and 0.5-10 wt% of a substance similar to 3-deoxy-D-manno-2-octulosonic acid (KDO). According to the present invention, the persimmon leaf-derived polysaccharide fraction promotes the production of interleukin-6 and TNF- to improve immune function, has no distinct toxicity to a macrophage, increases activity of a natural killer cell, and shows an effect of inhibiting spreading of cancer cells, thereby being effectively used to produce a composition for immunopotentiating or cancer-controlling food, a pharmaceutical composition for preventing or treating a disease caused by deteriorated immunity, or a pharmaceutical composition for preventing or treating cancer.
