Selective-plane illumination microscopy (SPIM) method for analyzing biological sample, involves changing brightness of image picked up by receiver during magnification change on drive unit
The method involves arranging a position-sensitive detector for detecting light emitted from the sample (3) in the detection direction. The magnification of the image field imaged on the detector is changed with a detector arranged in front of the zoom lens in the detection device. The brightness of the image picked up by the receiver is changed, and kept constant during the magnification change on a drive unit by adjusting the image contrast and/or the duration of the image recording and/or the illumination intensity in response to the set magnification change. An independent claim is included for a microscope and/or control unit for controlling exposure time, and illumination intensity, or image contrast for performing SPIM method.
