The method involves displacing a sample (3) for detecting different sample planes by a light sheet in direction of an optical axis of an imaging lens (4). The sample is continuously moved at constant speed through the sheet, and images are recorded at periodic time intervals by a detection device during movement of the sample. Image recording period or exposure time is measured in such a manner that a traverse path of the sample lies within a predetermined resolution region of the detection device. Speed of movement of the sample is determined by the period or the time. An independent claim is also included for a method for determining speed of movement of a sample in a selective-plane-illumination-microscopy.
