PROBLEM TO BE SOLVED: To provide a simple and effective method of synthesizing single-stranded DNAs, to provide a DNA chip made of the single-stranded DNAs prepared by the method and immobilized on a solid phase, and to provide a DNA synthesis kit.SOLUTION: A method of synthesizing single-stranded DNAs comprises: a process E of applying a reverse transcription reaction on a template RNA strand to synthesize a complex of the DNA strand and the RNA strand: and a process F of applying a heat treatment on the complex to dissociate into the RNA strand and the DNA strand, and decomposing the RNA strand by an RNase treatment.
