The present invention provides an isolated primate cell, preferably a human cell, comprising a genetically engineered disruption in a human leukocyte antigen (HLA) class II related gene that results in loss of MHC class II expression and function. The invention also provides an isolated cell further comprising a genetically engineered disruption in the β2 microglobulin (B2M) gene resulting in an HLA class I / class II deficiency. Also provided are methods of using the cells for transplantation and treatment of disease conditions.
