The present invention relates to detection of a target nucleic acid sequence using PTO cleavage and extension-dependent signaling oligonucleotide hybridization (PCE-SH). The present invention does not use a probe that hybridizes with a target nucleic acid sequence to provide a target signal. The present invention uses a probe (signaling oligonucleotide) that hybridizes with an extension strand formed in a target-dependent manner, and the extension strand is synthesized using CTO having an artificially selected sequence as a template.
