The present invention relates to: a culture medium composition including RGMc as an active ingredient for inducing a stem cell; and a method for inducing a stem cell using the same. RGMc is bound with neogenin and can produce an ICM cell expressing a stem cell genes such as OCT3/4, SOX2 and Nanohog in a blastocyst. The production level of the ICM cell is higher in a mouse embryo in which neogenin is over-expressed than in a normal mouse embryo, which is not treated with RGMc. Therefore, RGMc can be advantageously used for the culture medium composition for inducing the stem cell by using the same.
