A METHOD FOR EVALUATION OF WILD TYPE AND MUTANT TYPE ISOCITRATE DEHYDROGENASE ACTIVITY USING 13C LABELED -KETOGLUTARATE AND NUCLEAR MAGNETIC RESONANCE SPECTROSCOPY
The present invention relates to a method for evaluation of wild type and mutant type isocitrate dehydrogenase activity using 13C labeled -ketoglutarate and nuclear magnetic resonance spectroscopy to selectively monitor only -KG and 2-HG of cytolysate. According to the present invention, only -KG and 2-HG of many compounds of HDF cytolysate can be selectively monitored using 13C labeled -KG, thereby confirming presence or absence of IDH wild type and mutant type, enabling observation of enzyme activity, and being helpfully used for development of an enzyme inhibitor. Moreover, the developed inhibitor may be used a therapeutic agent for brain glioma.
