The present invention relates to a Luc 2 reporter vector inserted with a restriction enzyme cutting site into an open reading frame (ORF) of Luc 2, and a host cell induced with the vector. In addition, the present invention relates to a method for confirming 5 flanking region activation of an experiment target gene using the host cell. The Luc 2 reporter vector of the present invention can be usefully used in identifying and analyzing hexane regions which are important in gene control in the 5 flanking region of the experiment target gene by binding to the 5 flanking region of the experiment target gene.
