The present invention relates to a method for detecting specifically vancomycin-resistant Enterococci by using vancomycin-resistant Enterococci-specific primers and probes, and a test kit using the same. According to the method of the present invention, vancomycin-resistant Enterococci can be detected with very high efficiency by multiplex real-time polymerase chain reaction (PCR) using target genes (particularly, vanA and van B which are vancomycin-resistant genes), a D-alanine-D-alanine ligase (DDL) gene of Enterococci, and 16S rRNA-specific primers and probes. In addition, the kit of the present invention can conveniently and efficiently detect the target genes in a sample by the real-time PCR. Therefore, infection of the sample by vancomycin-resistant Enterococci can be detected in an easy manner by the method and the kit of the present invention. The method and the kit of the present invention can be more accurately applied to treatment of diseases.
