The present invention relates to a method of preparing 1,3-propanediol by using a mutant microorganism in which a 2,3-butanediol synthetic gene is deleted. More particularly, the present invention relates to a mutant microorganism having an ability to produce 1,3-propanediol from glycerol, wherein a gene for coding lactate dehydrogenase and a gene of an enzyme involved in synthesizing 2,3-butanediol are deleted, and a gene for coding pyruvate decarboxylase and a gene for coding aldehyde dehydrogenase are inserted or amplified, and to a method of promoting the production of 1,3-propanediol while suppressing the production of 2,3-butanediol by using the mutant microorganism. By using a glycerol-fermenting mutant microorganism according to the present invention, it is possible to remarkably improve the production amount of 1,3-propanediol while maximally suppressing the production of 2,3-butanediol.
