The present invention relates to genetically encoded FRET-based biosensors for monitoring the activity of matrix metalloproteinase 9 (MMP-9). MMP-9 is an extracellular endopeptidase involved in both physiological and pathological processes. Genetically encoded FRET biosensors anchored to the cell membrane make it possible to study the proteolytic activity of MMP-9 with high spatiotemporal resolution in the exact range of activity of MMP-9 in cells. The applicability of the biosensor both in vitro and in vivo in living cells was demonstrated by ratiometric analysis of biosensor cleavage with purified self-activated MMP-9.
