PROBLEM TO BE SOLVED: To provide a novel method of detecting target nucleic acid sequences on a solid phase using dual-labeled immobilized probes and its resistance to a 5'→3' exonuclease activity of a DNA polymerase.SOLUTION: The present invention requires no consideration of suitability of the position of a label to remain on a solid substrate because the label remains on the solid substrate due to resistance against a nuclease obtained by labeling a base component of an internal nucleotide. The present invention minimizes background signal by positioning the label at a site on a probe suitable to maximize quenching efficiency of a dual label system, since it can freely determine the position of an internal label on the probe.SELECTED DRAWING: Figure 1
