The various embodiments of the present invention provide a method for development of BYVMV resistant trait specific markers in okra. The BYVMV resistant and susceptible varieties are selected. The DNA is isolated from all susceptible and resistant okra varieties and subjected to agarose gel electrophoresis. Two resistant and two susceptible Okra plant genomes are subjected to RAPD profiling to analyze trait specific loci to select the polymorphic DNA marker. Total eight candidate markers are selected and screened in all susceptible and resistant varieties. Four markers are amplified and gel extracted for cloning and identification of sequences. The sequencing data is analyzed in NCBI database to determine the identity of sequences. The SCAR markers are designed for each locus based on the sequence obtained from all the polymorphic bands. The SCAR markers are amplified with nine susceptible and nine resistant varieties. 10 Claims, 11 Drawing Sheets
