METHOD FOR IDENTIFYING INTERSPECIFIC AND INTRASPECIFIC RELATIONSHIPS BETWEEN LACTIC ACID BACTERIA LACTOBACILLUS CASEI GROUP

PROBLEM TO BE SOLVED: To provide a degenerate primer for amplifying partial fragments of nucleotide sequences of mutL gene and dnaJ gene of lactic acid bacteria in the Lactobacillus casei group, and to provide a method and a kit, which discriminate interspecific and intraspecific relationships of lactic acid bacteria in the L. casei group.SOLUTION: The method according to the present invention extracts a DNA sample from lactic acid bacteria. Then, the DNA sample is amplified using a primer pair having a specific nucleotide sequence to obtain a partial fragment of a nucleotide sequence of a DNA mismatch repair protein (mutL) gene and/or a partial fragment of a nucleotide sequence of heat shock protein (dnaJ) gene. The sequence of the amplified partial fragment sequence is determined. The determined sequence is then compared with the corresponding gene sequence of two or more reference strains of Lb. casei group to identify the relative taxon of lactic acid bacteria.SELECTED DRAWING: None
