The present invention relates to an improved conjugation method for obtaining synthetic oligosaccharide / protein (OS / PR) conjugates. The method of synthetic OS / PR conjugate is a rapid method that provides a highly immunogenic, oligosaccharide / protein conjugate that elicits a specific and homogeneous immune response. Synthetic oligosaccharides comprise 4-8 repeat units of each monomer and at least one built-in terminal amino linker, such synthetic polysaccharides as Neisseria meningitidis serotypes A, C, Y, W, X and Haemophilus influenzae Mimics natural polysaccharides obtained from various Gram-negative bacteria, carrier proteins are obtained from Gram-positive bacteria such as Clostridium tetani (tetanus toxoid) or Corynebacterium diphtheriae (CRM197) or recombinant forms thereof. The conjugation chemistry of the oligosaccharide / protein conjugate of the present invention is a thioether bond. The present invention requires complete process time in the range of 14-22 hours.
