The present invention relates to an improved method of conjugation for the preparation of synthetic conjugates of oligosaccharide with protein (OS-PR). OS-PR synthetic conjugation method is a fast method that provides oligosaccharide-protein conjugates that are highly immunogenic and elicit specific and uniform immune responses. The synthetic oligosaccharide contains four to eight repeating structural units of the respective monomers and at least one embedded terminal amine linker, and this synthetic polysaccharide mimics the natural polysaccharide obtained from such gram-negative bacteria as serogroups A, C, Y, W, X Neisseria meningitidis and Ha influenza, and a carrier protein derived from gram-positive bacteria such as Clostridium tetani (tetanus toxoid) or Corynebacterium diphtheriae (CRM197) or their recombinant variants. The conjugation chemistry of said oligosaccharide protein conjugate according to the present invention is a thioester bond. The present invention provides for a complete process time in the range of 14-22 hours. These oligosaccharide protein protein conjugates are applicable in the preparation of a monovalent vaccine or a multivalent combination vaccine and as a diagnostic tool.
