PROBLEM TO BE SOLVED: To provide an OPA amino acid prelabeling analysis method capable of suppressing reduction in a column lifetime while suppressing the decomposition of an OPA labeled amino acid sample and also detecting a good peak.SOLUTION: The prelabeled amino acid analysis method by liquid chromatography includes a step of introducing an acid buffer, gas or a liquid having a pH of 6 to 7 and a sample solution including labeled amino acid and having a pH of 9 or more into a column filled with a silica gel filler in this order. It is preferable that a ratio (Va/Vs) of the volume (Va) of the acid buffer to the volume (Vs) of the sample solution is 0.5 to 1, a ratio (Vg/Va) of the volume (Vg) of the gas to the volume (Va) of the acid buffer is 1 to 2, and a ratio (Vl/Va) of the volume (Vl) of the liquid having the pH of 6 to 7 to the volume (Va) of the acid buffer is 0.5 to 1.
