摘要

A novel virus-like particle expression vector pTMSCA2C is provided. Firstly, apply a gene mutation technique to a plasmid pTrcHis-MS2, so as to mutate the fifth base T in the 19mer pack site genetic sequence of a MS2 bacteriophage into C to obtain a plasmid pTMSC; mutate the amino acid corresponding to the start codon of the maturation protein of a coding MS2 bacteriophage on the plasmid pTMSC from valine into methionine, in order to obtain a plasmid pTMSCA; and finally, apply a gene cloning technique to remove the stop codon from the genetic coding sequence of capsid protein of a wildtype MS2 bacteriophage, and then use the same to form a tandem array with the genetic coding sequence of a capsid protein of a His-tagged MS2 bacteriophage from a pseudo viral vector pTrcMS, and insert the genetic sequences in tandem into the plasmid pTMSCA to construct and obtain an expression vector pTMSCA2C.
