The present invention relates to a label-free nano-biosensor directly detecting C-reactive protein by using a reduced graphene oxide-nanoparticle mixture impedance measuring element and a method for directly detecting C-reactive protein without a label. According to the present invention, an ITO transformed mixture electrode in which a reduced graphene oxide and a gold nano-particle complex are deposited by chronoamperometry is used as a nano-biosensor. An antibody on the C-reactive protein of a person is fixed to the ITO transformed mixture electrode, and the C-reactive protein can be detected in low concentration. The detection method can specifically detect the C-reactive protein which is a target and can react to the C-reactive protein by being linearly proportional to a log scale concentration (from 1 ng mL^-1^ to 1000 ng mL^-1^) of the C-reactive protein in a solution. A detection limit reaches 0.06 or 0.08 ng mL^-1^, and an effect of obtaining the nano-biosensor with the high sensitivity on the C-reactive protein can be confirmed.;COPYRIGHT KIPO 2017
