Described are engineered strains of the oleaginous yeast Yarrowia lipolyticacapable of producing an oil comprisinggreater than 50 weight percent of eicosapentaenoic acid ["EPA"], an .omega.-3polyunsaturated fatty acid, measured as a weightpercent of total fatty acids ["% TFAs"] and having a ratio of at least 3.1 ofEPA % TFAs, to linoleic acid, measured as % TFAs.These strains over-express at least one .about.9 elongase/.about.8 desaturasemultizyme, in addition to other heterologous .about.9 elongases, .about.8desaturases, .about.5 desaturases, .about.17 desaturases, .about.12desaturases, C16/18 elongases, and optionally over-express diacylglycerolcholinephosphotransferases, malonyl CoA synthetases and/or acyl-CoAlysophospholipid acyltransferases. The expression of atleast one peroxisome biogenesis factor protein is down-regulated. Methods forproducing EPA within said host cells, oils obtainedfrom the cells, and products therefrom are claimed.
