The present invention relates to a cancer targeting drug delivery system comprising an indomethacin guided drug delivery conjugate (IGDDC). According to the present invention, provided is a novel cancer targeting drug delivery system having a high selectivity for cancer cells as compared with normal cells and being activated stepwise by histone diacetylase (HDAC) and cysteine cathepsin L (CTSL) to have an enhanced drug release efficiency, thereby suggesting a new approach to the development of a broad range of small molecule cancer chemotherapeutic agents.
