The present invention relates to a method for screening a substance which inhibits binding between mitsugumin 53 (MG53) and Orai1. According to the method of the present invention, it is possible to screen the substance capable of inhibiting binding between MG53 and Orai1, and the screened candidate substance increases activities of ryanodine receptor 1, enabling establishment of a screening system to develop treatment agents for various muscle diseases including hypotonus. Thus, the method can be widely applied to pharmaceutical industries.
