The present invention relates to a recombinant vector comprising a cellulolytic domain 3 and to a method for separating a target protein using the recombinant vector, wherein the method for separating the protein of the present invention binds the plant transformed with the recombinant vector to cellulose to easily isolate a fusion protein containing the target protein, and treats the fusion protein with enterokinase to efficiently isolate the target protein from the cell binding domain, thereby being expected to be applicable to various industrial fields.
