The present invention relates to a base sequence of a novel cellulose-binding domain; a recombinant vector comprising the same; and a protein isolation and purification method using the same. More specifically, the present invention relates to: a base sequence of a cellulose-binding domain, in which the 38th amino acid of family 9 carbohydrate-binding module (CBM9) domain of xylanase 10A protein from Thermotoga maritima, asparagine, is substituted with glutamine, and which encodes the cellulose-binding domain CBM9 (N38Q); a recombinant vector comprising the base sequence of the cellulose-binding domain; a peptide of a cellulose-binding domain, in which the 38th amino acid of CBM9 domain of xylanase 10A protein from Thermotoga maritima, asparagine, is substituted with glutamine, and which comprises an amino acid sequence of the cellulose-binding domain CBM9(N38Q); and a method of isolating proteins containing the cellulose-binding domain.
