The present invention relates to a method for identifying vector contamination during sequencing. The present inventors propose Vecuum as a technique for identifying the above-mentioned vector contamination. Vecuum not only gives better results than previous methods, but it can also reduce computation time for vector identification problems. Vecuum is also able to accurately identify contaminated genomic regions and detect most false variants that were not possible with previous techniques. In addition, we have applied known sequence information to Vecuum to identify a variety of external contaminants, including mammalian expression vectors, mouse genes in xenotransplantation, and prepped mRNA (cDNA) libraries. As a result, Vecuum will improve the reliability of low frequency somatic mutations in NGS sequencing data by providing a new quality control method for external contamination.;
