The present invention relates to a method for determining the content of an enzymatic entity E and the content and accessibility of one or more active sites of entity E in a liquid L comprising the steps of contacting L with a first binding moiety A1 that binds to a first binding site B1 of E that is different from the ligand binding site B2, and with a second binding moiety A2 that binds to B2. Furthermore, the present invention refers to a composition and a kit for use in the context of such method.
