Provided are non-human animals and methods and compositions for making and using the same, wherein the non-human animals comprise a mutant L-kynurenine hydrolase (or kynurenine) gene. The non-human animal is, in some embodiments, an amino acid that results in the removal of an epitope in the kynureninase polypeptide that is present in the membrane proximal outer region of human immunodeficiency virus-1 gp41. It may be described as having a genetic modification in an endogenous kynureninase gene to express a kynureninase polypeptide containing a substitution.
