Disclosed is a device for the automatic downhole connection of a built-in monitoring and adjustment cable of a string for intelligent separate layer water injection. The device comprises a water injection string (1), wherein a cable-releasing mechanism is arranged inside the water injection string, and the cable-releasing mechanism comprises a cable (7), a female cable joint (8), an electric motor (9) and a stressed blade (10). The female cable joint comprises a female joint housing (20), a jack insulation plate (21) and an electromagnet (22). A male cable joint (29) is further fixedly arranged inside the water injection string, and the male cable joint comprises a male joint housing (30), a contact pin insulation plate (31) and a neodymium-iron-boron magnet (32). A contact pin wire soldering point (36) is electrically connected to a sensor on an intelligent water distributor (4), and a wire (37), located in the contact pin insulation plate, is connected between a contact pin (35) and the contact pin wire soldering point. Further disclosed is a method for the automatic downhole connection of a built-in monitoring and adjustment cable of a string for intelligent separate layer water injection by the automatic connection device. The automatic connection device has the advantages of the male and female joints being firmly connected, being able to avoid short-circuiting and wet-plugging underground, high connection efficiency, and simple operations, and can realize real-time communication between the ground and the underground.
