Provided is a monoclonal antibody that specifically binds human plasmalemma vesicle-associated protein (PLVAP, shortly PV-1) or a derivative thereof, wherein same comprises the amino acid sequences of SEQ ID NO: 17, SEQ ID NO: 18 and SEQ ID NO: 19 of the antigenic complementarity determining regions CDR1, CDR2 and CDR3 in the light chain variable region of the antibody; and the amino acid sequences of SEQ ID NO: 22, SEQ ID NO: 23 and SEQ ID NO: 24 of the antigenic complementarity determining regions CDR1, CDR2 and CDR3 in the heavy chain variable region of the antibody, respectively. Also provided are a preparation process of a human-mouse chimeric antibody of the antibody and the amino acid sequences of the heavy chain variable region and the light chain variable region of the human-mouse chimeric antibody. The antibody or derivative thereof can serve as a component of a pharmaceutical composition or can be prepared into an appropriate drug preparation, administered alone or in combination with other drugs such as an anti-VEGF monoclonal antibody and the like, for treating choroidal neovascular fundus oculi diseases and other angiogenesis/osmosis-related diseases.
