This disclosure relates to a method of treating cancer by delivery in a tumor of a tumor containing a Toll-like receptor 9 agonist (TLR9) and a tumor antigen, wherein the TLR9 agonist is a polynucleotide or a chimeric compound thereof. The methods of the present disclosure involve injecting particles in at least one dose and are effective in treating both injected and uninoculated tumors of a mammalian subject. Additionally, the present disclosure provides immunogenic compositions containing particles as well as methods of making the same.
