The present invention relates to an L-arabinose-based L-ribose manufacturing method. More specifically, the manufacturing method of L-ribose comprises: a step of culturing a first microorganism with an L-ribose synthetic pathway introduced from L-arabinose on a medium including arabinose to produce L-ribose; and a step of removing the first microorganism from the culture medium, then inoculating a second microorganism with a metabolic pathway introduced thereto to culture the second microorganism to consume L-arabinose and L-ribulose, and then recovering the produced L-ribose. The L-ribose manufacturing method uses all saccharides except L-ribose in metabolism to remove saccharides. Therefore, high-purity L-ribose can be produced from fermented products. The produced ribose can be effectively used as a synthesis starting material of various L-type nucleotide sugar medicines when manufacturing medicines.
