The present invention relates to a method for increasing read data analysis accuracy by removing primer sequence information present in a read obtained through a next generation sequencing (NGS). More specifically, the present invention relates to a method for determining the primer sequence information in the read by matching the read and designed primer information to various reference values over various stages, and then correcting only the primer sequence, thereby increasing the accuracy of the Read data analysis. According to the present invention, the method for increasing read data analysis accuracy in amplicon based NGS by using primer remover has fast data analysis speed and only the primer sequence can be precisely removed so that is useful for the increasing the efficiency and accuracy of read data analysis.
