The present invention relates to a method for the preparation of isoquercetin and α-glycosyl isoquercetin. Specifically, the present invention relates to a method for economically preparing α-glycosyl isoquercetin widely used in food and cosmetic fields as an antioxidant, a color stabilizer, a fragrance change inhibitor, an ultraviolet absorber, and a body fat reducer, by using isoquercetin, high purity of which is prepared by allowing a reaction to proceed while removing free rhamnose from the reaction system, in preparing isoquercetin from rutin by an enzymatic method.
