The present invention relates to a method of forming a salivary gland organoid through three-dimensional stem cell culture, and more particularly, to a method of regenerating a salivary gland by co-culturing adult salivary stem cells and vascular endothelial stem cells isolated from human salivary glands (Organoids) of the like. When the salivary stem cells and the vascular endothelial stem cells were co-cultured, the organoid was observed, and it was confirmed that the organoid was formed without being affected by the cell proliferation. The organoid was found to be involved in disease modeling, pathological studies, drug screening, Toxicity assessment, and genetic engineering.
