The present invention relates to a method for generating salivary gland organoids through 3D sequential culture of human salivary gland stem cells and a use of salivary gland therapy using the same. Specifically, the present invention relates to the method for generating the salivary gland organoids having characteristics of various salivary gland cells, primarily by enhancing a function of the organoids through 3D spheroid culture of the human salivary gland stem cells and by 3D sequential culture of the organoids. The present inventors have verified that separating conditions of a single tissue stem cell from the human adult salivary gland and 3D culture conditions in which homogeneous organoids are generated can be determined to generate salivary gland tissue-simulated organoids. The organoids can be used for disease modeling, pathology studies, drug screening, toxicity evaluation, and genetic engineering.
