The present invention relates to a protein comprising the protruding domain of the Japanese encephalitis virus coat protein, a homodimer thereof, and a pharmaceutical composition for preventing or treating a disease caused by the Japanese encephalitis virus infection comprising the same as an active ingredient. The present invention utilizes an insect cell expression system to provide homodimeric protein antigens of structurally stable overhang domains as originally present on the surface of a virus. In addition, the antigen may be used as a vaccine more stable than the conventional Japanese encephalitis vaccine because of its excellent ability to produce antibodies in the subject when injected into the subject, and may be used for diagnosis of Japanese encephalitis.
