The present invention provides compositions and methods for treating or preventing pain. The provided compositions are resistant to overdose and abuse. Provided compositions comprise at least one GI enzyme-sensitive opioid, comprising two or more different molecules, each molecule comprising an opioid agonist covalently linked to at least one GI enzyme inhibitor subunit. Includes agonist-releasing subunits. The compositions described herein can achieve plasma profiles that are distinct from compositions that include one similar polysubunit molecule.
