To provide production methods for a mesenchymal cell composition by separating amnion-derived MSC of high purity through only one enzymatic treatment simply and aseptically.SOLUTION: Provided are a production method for a mesenchymal cell composition and a production method for a cryopreserved mesenchymal cell composition which comprise applying an enzyme treatment to amnion by collagenase, and thermolysin and/or dispase; and passing the enzymatically treated amnion through a mesh, as well as a therapeutic agent for graft-versus-host disease, inflammatory bowel disease, systemic lupus erythematosus, liver cirrhosis, and radiation enteritis which contains the mesenchymal cell composition as an active ingredient.SELECTED DRAWING: None
