The present invention relates to a method of tagging DNA-encoded chemical libraries. The method comprises binding a first functional group of a bifunctional linker to a headpiece comprising an initiator oligonucleotide, wherein said initiator oligonucleotide comprises a hairpin structure; binding a second functional group of the bifunctional linker to a first building block; reacting a second building block with said first building block in an organic solvent; and binding an identifier region at the 3' end of said headpiece, wherein the identifier region encodes said second building block. The method provides a step-wise approach to screening and identifying small molecules that have greater potency.
