The present invention relates to a method of diagnosing premature ovarianfailure in a femalesubject and a method of detecting a decline in non-atretic follicles in afemale subject,comprising detecting a decrease in the level of Vasa expression in a sample ofperipheralblood obtained from the female subject by comparing it with the level of Vasaexpression inthe peripheral blood of a female subject having a functional reproductivesystem. Thepresent invention also relates to a method of detecting a fluctuation in thenumber ofcirculating female germline stem cells in a female subject, said methodcomprisingcomparing the level of Vasa expression in a sample of peripheral bloodobtained from thefemale subject at a first time point to the level of Vasa expression in asample obtained fromthe female subject at a second time point.
