The present invention provides a method for producing sialic acid modified N-glycosylation recombinant protein in a recombinant escherichia coli periplasmic space. In particular, the present invention relates to producing sialic acid modified N-glycosylation recombinant protein in a recombinant escherichia coli strain W3110ΔnanKETA::Kan body, to achieve sialic acid modified N-glycosylation recombinant protein in a escherichia coli periplasmic space, wherein a sialylated oligosaccharide chain is Neu5Ac-α-2,6-Gal-β-1,4-GlcNAc-β-1,3-Gal-β-1,3-GlcNAc. The method is short in time, high in efficiency, and low in cost, does not require the addition of exogenous sialic acid, and establishes a technology platform for the production of therapeutic sialic acid modified N-glycosylation protein drugs.
