The present invention relates to a manufacturing method for an IsPETase protein crystal, a method for screening an IsPETase protein activity regulator by using a 3D structure of the protein, a method for screening an IsPETase variant, an IsPETase variant with improved PETase activity, and a PET decomposition method using same. The present invention specifies the method for effectively manufacturing an IsPETase protein crystal, and provides an acquired crystal manufactured thereby. In addition, the manufacturing method specifies the tertiary structure of IsPETase from the crystal, and manufactures a variant with improved PETase activity based on the structure. Therefore, the IsPETase variant can be effectively utilized in the PET decomposition area.
