The present invention relates to a kit for detecting a target substance comprising two segments divided from G-quadroplex. The present invention also relates to a method for determining the presence of a target substance comprising the step of treating the kit with a sample. The present invention can easily and quickly detect a target substance by using fluorescence signal differences of fluorescent base analogs present in the divided G-quadroplex DNA, and can be utilized for detecting various biological materials and chemicals.
